Intended use:

Salmonella detection by real-time PCR

 

The test detects Salmonella DNA.
Each reaction contains an internal amplification control (PLUS). The real-time PCR assay can be used with
established real-time PCR instruments, equipped for detection of two fluorescence emissions in the FAM and
VIC/HEX channel at the same time. The technical validation of instruments was performed on Agilent
Mx3005P, BioRad CFX 96, Roche LightCycler® 480 II, Roche LightCycler® 2.02, Applied Biosystems 7500,
Qiagen RotorGene Q and Cepheid SmartCycler®.

General Information:

Salmonella is a genus of rod-shaped Gram-negative enterobacteria that causes typhoid fever, paratyphoid fever and foodborne illness. It is motile in nature and produces hydrogen sulfide.

Disease-causing salmonellae have recently been re-classified into a single species, Salmonella enterica, which has numerous strains or serovars. Other salmonellae are frequent causes of foodborne illness, and can especially be caught from poultry and raw eggs and more generally from food that has been cooked or frozen, and not eaten straight away. In March 2006, the U.S. government reported that 16.3% of all chickens were contaminated with salmonella. In the mid to late 20th century, Salmonella enterica serovar Enteritidis was a common contaminant of eggs. This is much less common now with the advent of hygiene measures in egg production and the vaccination of laying hens to prevent salmonella colonisation.

Sample Preparation For DNA-preparation the use of SureFood® PREP Salmonella kit is recommended.
To assess results in matrices with low or zero microbial activity, it is recommended to compare the Cp-value
at the beginning and at the end of the pre-enrichment (dynamic).
Detection Limit The SureFood® PATHOGEN Salmonella PLUS real-time PCR has a limit of detection of ≤ 5 DNA copies. The
assay limit of detection depends on sample matrix, processing grade, DNA preparation and DNA content.
