Intended use:

The test detects pork DNA (Sus scrofa). Each reaction contains an internal amplification control and an internal detection assay for animal DNA (IAAC).

For DNA-preparation of raw material the use of SureFood® PREP Basic and for highly processed food and feed the use of SureFood® PREP Advanced is recommended.

General information:

The real-time PCR assay can be used with established real-time PCR instruments, equipped for detection of two fluorescence emissions at 522 nm and 553 nm at the same time. The technical validation of instruments was performed on Roche LightCycler® 2.0*, Roche LightCycler® 480 II, Qiagen Rotor-Gene Q, Applied Biossystems 7500, Bio-Rad CFX96 and Agilent Mx3005P.

*note: A Color Compensation is necessary for users of the Roche LightCyclerR 2.0. The SureCC Color Compensation kit II (Art. No. F4010) must be used for the color compensation of such devices.

SureFood® ANIMAL ID Pork Sens PLUS

Intended use:

The test detects pork DNA (Sus scrofa). Each reaction contains an internal amplification control.

General information:

The real-time PCR assay can be used with established real-time PCR instruments, equipped for detection of two fluorescence emissions at 510 nm and 580 nm (FAM and VIC/HEX) at the same time. The technical validation of instruments was performed on Agilent Mx3005P, Agilent AriaDx, BioRad CFX 96, Roche LightCycler® 480 II, Roche cobas z 480 Analyzer, Roche LightCycler® 2.02, Applied Biosystems 7500, LTF MyGo Pro, BMS MIC and Qiagen RotorGene Q.

Comment:

Many conventional produced meat products show an unavoidable cross contamination when pork and beef or poultry is produced on a shared production line. Although not detectable with common systems, these contaminations can be detected by using SureFood® ANIMAL ID Pork SENS. Currently there is no internationally harmonized threshold for a non declaration of pork content of food. In many European countries an undeclared pork amount of 0.5 to 1 % or less is mostly accepted by the governmental authorities (the detection limit of SureFood® ANIMAL ID Pork SENS is far below that).

In other region of the world different acceptable level of contamination might be accepted. These countries must find practically acceptable thresholds for pork content since unavoidable contaminations can be detected with novel analytical methods like SureFood® ANIMAL ID Pork SENS.

The test contains an internal inhibition control (PLUS) for detection in the V / R /  LC detection channel.

Accessories:

For Roche Lightcycler LC1.5 und LC2.0 device, a colour compensation with article S4010 is necessary.

SureFood® Animal QUANT Pork

Intended use:

The test detects the relative quantitative pork DNA amount in meat samples. Therefore the kit contains two PCR systems, one for detection of a pork specific gene (Sus detection gene) and one for the detection of an animal gene (Ref reference gene).

For DNA-preparation of meat samples the use of SureFood® PREP Basic  is recommended.

General information:

The real-time PCR assay can be used with established real-time PCR instruments (Roche LightCycler®, Rotor-Gene Q, ABI PRISM, Eppendorf realplex, BioRad CFX96, Agilent MxSeries etc.).

Detection limit The pork specific PCR has a limit of detection of ≤ 5 DNA-copies. The assay limit of detection depends on sample matrix, processing grade, DNA-preparation and DNA-content.
The limit of quantitation depends on the concentration of the sample DNA used in the analysis. Forexample, if 50,000 target-sequence copies of the reference gene are present, the relative quantitation limit for pork DNA is 0.04 %.

The test detects pork (Sus scrofa), chicken (Gallus gallus) and turkey (Meleagris gallopavo) DNA. Each reaction contains an internal amplification control and an internal detection assay for animal DNA (IAAC).

For DNA-preparation the use of SureFood® PREP Basic and for highly processed food and feed the use of SureFood® PREP Advanced is recommended.

General information:

The real-time PCR assay can be used with established real-time PCR instruments, equipped for detection of four fluorescence emissions at 522 nm, 553 nm, 610 nm and 670 nm (FAM, VIC, ROX and Cy5) at the same time. The technical validation of instruments was performed on Roche LightCycler® 480 II*, Roche cobas z 480 Analyzer*, Qiagen Rotor-Gene Q, Applied Biosystems 7500, Bio-Rad CFX96 and Agilent Mx3005P.
