Intended use:

The test detects horse DNA (Equua caballus). Each reaction contains an internal amplification control and an internal detection assay for animal DNA (IAAC).

For DNA-preparation of raw material the use of SureFood® PREP Basic and for highly processed food and feed the use of SureFood® PREP Advanced is recommended..

General Information:

The real-time PCR assay can be used with established realtime PCR instruments, equipped for detection of two fluorescence emissions at 522 nm and 553 nm at the same time. The technical validation of instruments was performed on Roche LightCycler® 2.0*, Roche LightCycler® 480 II, Qiagen Rotor-Gene Q, Applied Biossystems 7500, Bio-Rad CFX96 and Agilent Mx3005P.

*note: A Color Compensation is necessary for users of the Roche LightCyclerR 2.0. The SureCC Color Compensation kit II (Art. No. F4010) must be used for the color compensation of such devices.

The test detects horse DNA (Equus caballus) and donkey DNA (Equus asinus) seperately. Each reaction contains an internal amplification control and an internal detection assay for animal DNA (IAAC).

For DNA-preparation the use of SureFood® PREP Basic  is recommended.

The real-time PCR assay can be used with established real-time PCR instruments, equipped for detection of three fluorescence emissions at 522 nm, 553 nm and 670 nm. The technical validation of instruments was performed on Roche LightCycler® 480 II, Qiagen Rotor-Gene Q, Applied Biosystems 7500, Bio-Rad CFX96 Agilent Mx3005P.

IAAC= Identification Amplification Animal Control
The IAAC kit requires a 3 channel real-time thermocycler.

SureFood® ANIMAL QUANT Equus

Intended use:

The test detects the relative quantitative equine DNA. The family of Equidae contains horse, donkey and zebra. The kit contains two PCR systems, one for detection of an equine specific gene (Equus detection gene) and one for the detection of an animal gene (Ref reference gene).

For DNA-preparation the use of SureFood® PREP Basic  is recommended.

General Information:

The real-time PCR assay can be used with established real-time PCR instruments (Roche LightCycler®, Rotor-Gene Q, ABI PRISM, Eppendorf realplex, BioRad CFX96, Agilent MxSeries etc.).

Detection limit The equine specific PCR has a limit of detection of ≤ 5 DNA-copies. The assay limit of detection depends on sample matrix, processing grade, DNA-preparation and DNA-content.
The limit of quantitation depends on the concentration of the sample DNA used in the analysis. For example, if 50.000 target-sequence copies of the reference gene are present, the relative quantitation limit for equine DNA is 0.05 %.

The test detects beef (Bos taurus), horse (Equus caballus) and pork (Sus scrofa) DNA. Each reaction contains an internal amplification control and an internal detection assay for animal DNA (IAAC).

General Information:

The real-time PCR assay can be used with established real-time PCR instruments, equipped for detection of four fluorescence emissions at 522 nm, 553 nm, 610 nm and 670 nm (FAM, VIC, ROX and Cy5) at the same time. The technical validation of instruments was performed on Roche LightCycler® 480 II/Roche cobas z 480 Analyzer2, Qiagen Rotor-Gene Q, Applied Biosystems 7500, Bio-Rad CFX96, BMS MIC, Agilent Mx3005P and Agilent AriaDx.
