Lateral Flow Milk
Intended use:
The Lateral Flow Milk assay is a qualitative detection assay of Milk residues in surface samples got from e.g. production lines. The assay is also applicable for food samples. The assay is applied for the detection of small amounts of Milk contaminations.
General Information:
Milk contaminations in food possibly elicit hypersensitivity in sensitive persons. Food production lines have to be free of residues to prevent these allergic reactions. The effectiveness of cleaning can be verified by using the Lateral Flow Milk Swabbing method.

RIDASCREEN®FAST Milk
Intended use:
RIDASCREEN®FAST Milk is a sandwich enzyme immunoassay to quantify milk proteins in food containing whey, milk or milk powder such as sausages, ice cream, chocolate, bakery goods, cake and bread mix, soups, sauces, dressings and beverages (juice, wine, beer). The antibodies specifically detect caseins as well as β-lactoglobulin of cow’s milk and of sheep’s, goat’s and buffalo’s milk.
Approvals:
The ELISA RIDASCREEN®FAST Milk was approved by the AOAC Performance Tested MethodsSM Program and was assigned PTM Certification No. 101501 by the AOAC Research Institute. The in-house validation data was confirmed by an independent lab. The sandwich ELISA is applicable for the detection of milk protein (casein und b-lactoglobulin) in cookies, chocolate, ice cream, infant formula and sausages.

The assay results are quantitative if the sample contains milk or milk powder. If the milk protein composition of the sample is not known and the milk protein ratio is different than the natural ratio in milk (e.g. a sample primarily consisting of whey), the results may be underestimated. If a quantitative result for caseins and/or β-lactoglobulin is requested, the sample should be tested in the RIDASCREEN®FAST Casein (R4612) and/or the RIDASCREEN®FAST β-lactoglobulin (R4902).

General Information:
Cow`s milk contains 3.2 % proteins which consist of 10 % β-lactoglobulin (leading protein of whey) and 80 % caseins. The most important allergen is β-lactoglobulin especially for children while the caseins become to be dominant later in adults.
Casein (lat. Caseus = cheese) is a rough flaked curdling protein, which forms micells in the milk and precipitates under acidic conditions. The group of caseins consists of the αs-caseins, the β-caseins, the κ-casein, and the γ-caseins (proteolytic protein fragment of β-casein by the milk protease plasmin). Κ-casein is cleaved into a hydrophobic (para κ-casein) and into a water soluble polar component (macropeptid) by proteolysis, e.g. by using lab ferment.
The main components of whey are β-lactoglobulin and α-lactalbumin besides lactoferrin, bovine serum albumin and immunoglobulins.
Milk can be present as an ingredient or as a contamination in raw and processed food products. Because milk can induce allergic reactions at infancy it must be declared as an ingredient on food labels. Whey (β-lactoglobulin) or caseinates (e.g. in sausages) are often added to food products, therefore it is recommended to determine casein in food.

RIDASCREEN® β-Lactoglobulin
Intended use:
RIDASCREEN® β-Lactoglobulin is a competitive enzyme immunoassay for the quantitative analysis of β-lactoglobulin in hydrolyzed milk products including hypoallergenic baby food. The assay is calibrated to hydrolyzed β-lactoglobulin check samples; the standards consist of native β-lactoglobulin. The assay detects native and processed protein and fragments thereof (during the heating of milk, the native ß-lactoglobulin is partially denatured).

Reference Material
Certified reference material plays a decisive role in food analysis. But so far, suitable material was missing for allergen analysis. Now, MoniQA Association has presented the first validated reference material for the analysis of milk allergen.
For many laboratories, they are an important component of quality assurance: certified reference materials. They allow the user to assess whether the test results are reliable and to rule out false positive or false negative results. Only recently, we emphasized the importance of reference material. However, certified reference material is not available for every field of analysis. One major challenge is the production of reference material for allergen analysis. Allergenic proteins may alter and change their structure or concentrations – and thus cause instability and unconformity.
Allergen analysis is a field with a high demand for certified reference material. The most common technique for the detection of allergens is an ELISA test – a method which is very effective, easy to use and relatively cheap. Yet, comparability of measured results is difficult without reference material. Due to high demand, there have been continuing efforts to develop certified reference material also for allergen analysis. In 2013, the renowned MoniQA Association (International Association for Monitoring and Quality Assurance in the Total Food Supply Chain) launched a respective initiative.
Since January 2017, the first validated reference material for food allergens is available at R-Biopharm. The portfolio, produced by Trilogy Laboratories, includes four testing materials for milk allergen:
